Comparative Study on the Dissolution Rate of Acetaminophen from Sustained Release Dosage Form

Abstract
We have studied the dissolution kinetics of two sustained release tablets containing acetaminophen (Tylenol ER and Taspen ER) in five media, and the dissolution profiles were compared. Dissolution for the first 15 minutes from Tylenol ER was much faster than Taspen ER, indicating that the fast releasing component of the Tylenol ER rapidly dissolves in the medium. Rapid-dis-solving component of Tylenol ER, actually, dissolved and released drug completely in two minutes. However, there was no release of drug from Taspen ER for the first two minutes. Tylenol ER released over 90% of the drug in 3 hours in all mediums. However, it took about 6 hours for Taspen ER. Since the drug is for antipyretic/ analgesic effect, rapid dissolution can be advantageous. The comparison of the dissolution profile, based on the guideline of KFDA, indicates that the two tablets are not equivalent in their dissolution.
